Wayne County is a county located in the U.S. state of Mississippi. As of the 2020 census, the population was 19,779. Its county seat is Waynesboro. The county is named for General Anthony Wayne.

According to the U.S. Census Bureau, the county has a total area of 814 square miles (2,110 km2), of which 811 square miles (2,100 km2) is land and 2.8 square miles (7.3 km2) (0.3%) is water. It is the fifth-largest county in Mississippi by land area.
